The MIKE linebacker's job is to stop anything coming up the middle between the tackles as well as keep an eye on the whole field to watch the play develop. They will also be responsible for any second man coming out of the backfield on either side. They must be strong as well as very aggressive to fight off the big blockers. It is always good to have a Middle backer who is a little nuts.
The SAM linebacker's job is to protect against the run on the strong ( side w/ the TE) side as well as drop into coverage, usually in the curl to flat zones. They will typically cover the tight end in man-to-man coverage and must be strong to fight off any blocks from the TE or FB since running plays will usually go their way.
The WILL linebacker has many of the same duties as the SAM, but is on the other side. He is typically the fastest of the 3 because he is called into pass coverage the most. It is also important to be fast since they usually are pursuing the play from the backside. He will watch to contain the run on his side as well as drop into coverage on a pass play.

That's about it.
